Scope and Importance

Immunoprecipitation (IP) is the small-scale affinity purification of antigens using a specific antibody and is one of the most widely used methods for antigen purification and detection. This approach enables researchers to:identify the activation status of proteins, determine post-translational protein modifications, measure the molecular weight of a given protein, capture protein-binding molecules in the study of protein-protein and protein-nucleic acid interactions

Individual protein immunoprecipitation used for biochemical characterization of proteins involves the use of antibody that is particular for a known protein so as to isolate that particular protein out of solution. Chromatin immunoprecipitation is used to determine the location of DNA binding sites on the genome for a particular protein of interest. The advantage of chromatin immunoprecipitation is that protein–DNA interactions inside the nucleus of living cells or tissues can be identified. Protein complex immunoprecipitation also called as co-immunoprecipitation is based on the selection of the antibody that targets a known protein only so that by targeting this known protein with an antibody it may become possible to pull the entire protein complex out of solution and thereby unknown proteins in the complex can be identified.
